چکیده

Objective: A novel metalloproteinase, Pregnancy Associated Plasma Protein-A (PAPP-A), has been implicated in the development of atherosclerosis, and is under consideration as a novel biomarker for acute coronary syndrome and unstable plaque. The aim of this study was to determine if PAPP-A overexpression directly contributes to plaque vulnerability. Methods: Apolipoprotein E knock-out (ApoE KO) mice, a model of atherosclerotic development, and ApoE KO mice overexpressing PAPP-A transgene in arterial smooth muscle (ApoE KO/Tg) were fed a high fat diet for 20 or 40 weeks starting at seven weeks of age. At harvest, the brachiocephalic artery (BCA) was fixed, embedded in paraffin, sectioned and stained for morphologic analyses. Results: After 20 weeks on high fat diet, significantly more ApoE KO/Tg mice had BCA lesions with a necrotic core and fibrous cap than did ApoE KO mice. After 40 weeks on high fat diet all mice had BCA plaques with necrotic cores, but plaque progression with healed ruptures (i.e., buried fibrous caps), and plaque inflammation were greater in BCA of ApoE KO/Tg mice than in ApoE KO mice. Conclusion: Overexpression of PAPP-A in arterial smooth muscle of ApoE KO mice is associated with accelerated plaque progression and development of vulnerable and ruptured plaque.

Hepatic caveolin‐1 is enhanced in Cyp27a1/ApoE double knockout mice

Sterol 27-hydroxylase (CYP27A1) is involved in bile acid synthesis and cholesterol homoeostasis. Cyp27a1(-/-)/Apolipoprotein E(-/-) double knockout mice (DKO) fed a western diet failed to develop atherosclerosis. Caveolin-1 (CAV-1), the main component of caveolae, is associated with lipid homoeostasis and has regulatory roles in vascular diseases. We hypothesized that liver CAV-1 would contribu...

Lentiviral-mediated RNA interference of lipoprotein-associated phospholipase A2 ameliorates inflammation and atherosclerosis in apolipoprotein E-deficient mice.

Lipoprotein associated phospholipase A2 (Lp-PLA2) overexpression is implicated in athero-sclerosis. In the present study, we evaluated the effects of lentiviral-mediated RNA interference (RNAi) of Lp-PLA2 on inflammation and atherosclerosis in apolipoprotein E-deficient mice. Apolipoprotein E-deficient mice were randomly allocated to control and experimental ...

CD8+ T Cells Mediate the Athero-Protective Effect of Immunization with an ApoB-100 Peptide

Immunization of hypercholesterolemic mice with selected apoB-100 peptide antigens reduces atherosclerosis but the precise immune mediators of athero-protection remain unclear. In this study we show that immunization of apoE (-/-) mice with p210, a 20 amino acid apoB-100 related peptide, reduced aortic atherosclerosis compared with PBS or adjuvant/carrier controls. Immunization with p210 activat...

Overexpression of IL-18 decreases intimal collagen content and promotes a vulnerable plaque phenotype in apolipoprotein-E-deficient mice.

OBJECTIVE Although IL-18 has been implicated in atherosclerotic lesion development, little is known about its role in advanced atherosclerotic plaques. This study aims to assess the effect of IL-18 overexpression on the stability of preexisting plaques. METHODS AND RESULTS Atherosclerotic lesions were elicited in carotid arteries of apolipoprotein E (apoE)-deficient mice (n=32) by placement o...

RANTES/CCL5 and Risk for Coronary Events: Results from the MONICA/KORA Augsburg Case-Cohort, Athero-Express and CARDIoGRAM Studies

BACKGROUND The chemokine RANTES (regulated on activation, normal T-cell expressed and secreted)/CCL5 is involved in the pathogenesis of cardiovascular disease in mice, whereas less is known in humans. We hypothesised that its relevance for atherosclerosis should be reflected by associations between CCL5 gene variants, RANTES serum concentrations and protein levels in atherosclerotic plaques and...
